Arrhythmia can be produced by diverse groups of drugs, which prompted regulatory agencies to mandate preclinical testing of all pharmaceuticals to exclude unwanted side effects due to hERG channel inhibition. Until now, the hERG patch clamp assay has been the most valuable and reliable tool for assessing a potential drug-induced blockade of the hERG channel.
However, this preclinical assay addresses only a single cardiac target. Any drug interaction on other ion channels important for the formation of the cardiac action potential could also lead to severe cardiac arrhythmias.
